There are alot of music-based games out right now and more coming out in the future(DJ Hero exists, people; its coming), so this doesnt sound like much of a difference. If it is exactly like playing a real guitar(with strings instead of buttons), then those that actually play guitar IRL will be drawn more to it, whereas those that don't/can't play will not be drawn to it so easily. The insrtument/controls will determine how well people warm up to the game.
